Does a kicker or multi-kicker cost count as a spell?

Example: There is a card:Wurm's Tooth on the field. Play a green creature with multi-kicker. Kick it 6 times - Would one gain life for each time kicked on top of the 1 life for casting the spell?

Nooope. It's only one spell, regardless of how many times you kick it.

Nope, just that one spell that got kicked.

So, a Wolfbriar Elemental kicked 6 times would only trigger the tooth once.

Unless I'm horribly mistaken for some reason...

Kicking a spell occurs at the same time as you would choose the effect/s of a modal spell (Esper Charm , Austere Command ). It's a modification of the spell's effect, not the casting of a new spell/s.

Bonus Answer :

Even a card that uses the Replicate mechanic like Gigadrowse wouldn't trigger a card:Kraken's Eye more than once, because copies aren't cast, they're put into play/on the stack.

The trigger on the lucky charms are very specific in that the spell itself must be cast (although it doesn't need to resolve! Once the spell is 'cast', the charm trigger goes on top of it on the stack and will resolve before the spell itself does, meaning your opponent could counter the spell you cast and you'd still gain the life).
