does the opponent get a poison counter when you play Hand of the Praetors ?

Right. A permanent must be on the battlefield for it's ability to trigger.

No, as you are at the stage of casting the spell, not resolving.
so even if the infect creature spell being casted is countered, the effect will still go onto the stack and resolve. (assuming Hand of the Praetors is on the field)
